My visa is expiring in June and right now I'm applying for the 3-year extension. I have to travel to London for work sometime in April/May - anyone know if I'm able to come back into the states once I leave and come back before it expires? I just want to make sure I don't jeopardise my chances of getting back into the US.

It shouldn't matter as long as your current H-1B has not expired. You'll also need to double check that the visa stamp in your passport has not expired. If it has, you'll need to get a new visa stamp before you return to the country. Of course, this doesn't really make too much sense since you'll be getting an extension and your new visa stamp will only go until the end of your current visa status.
You should leave the country to get your extension anyway. Why not get your extension from the US embassy in London, or won't it be ready by that time?
I would ask an immigration lawyer if you have access to one, but I think you should be able to travel once you're back in the US before the last day that's written on the stamp on your I-94.
